Abstract: HCV entry is an unusually complex process, requiring numerous host entry factors. These host entry factors have distinct subcellular localizations with some localized to the basolateral membrane and others at the tight junction.

We combined single particle imaging of fluorescently labeled virions with the development of a polarized 3-dimensional organoid culture system to define the steps of HCV entry. The pathway and its potential implication for HCV biology will be discussed.
